Best possible answer is 2. Protein is in insoluble/pellet because its getting denatured at that particular temperature. Lowering temperature will prevent this and solublize the protein

Apologize. I used the Set A question paper. Image did not load.

I think the picture below summarizes. If it had a correct orientation(1st plasmid) - BamHI + HindIII  == 0.5 + 4.5
                                                                                                                              PstI    + HindIII   == 1.5 + 3.5
Answer is C. It will be reversed(A) for incorrect orientation.

Key for search: Circular Dichroism, psychrophile, thermophile, protein....

May be the answer is : d.. bcz of salt bridge made of by the hydrogen bond and we knw H- bonding highly stable...for that reason last one is the answer...what is the answer???
